Effect of siRNA-mediated knockdown of p22phox on the phosphorylation of ERK1/2 and JNK by lysoPC. HUVECs were transfected with either nonsilencing control siRNA (n.s.RNA) or p22phox siRNA (detailed in Materials and Methods). At 48 hours after transfection, cells were serum-starved for 16 hours and stimulated with lysoPC (30 μmol/L) for 15 minutes. A: Western blot analysis shows almost complete disappearance of p22phox in cells treated with p22phox siRNA compared with control or n.s.RNA-treated cells. The cell lysates of naïve and retinoic acid-treated HL60 cells were used as an indicator of the p22phox relative electrophoretic mobility.
