ClpP-dependent degradation of ClpE. A strain expressing clpP under the control of the xylose-inducible promoter Pxyl was grown in the absence or presence of xylose. Cells were grown exponentially in the absence of ClpP in LB medium at 37°C. At an OD540 of 1.0, the first sample was taken without the addition of xylose (− xylose). After 30 min, xylose was added to the culture (final concentration, 0.5%) in order to induce the synthesis of ClpP; samples were taken after 0 min, 1 h, and 2 h (+ xylose). Twenty-five micrograms of total protein was loaded per lane. ClpE and ClpP were detected with specific antisera.
